About Capstan TherapeuticsCapstan Therapeutics, Inc. is a privately held biotechnology company dedicated to advancing in vivo cell reprogramming. The platform technology has the potential to generate transformative therapies with possible applications across a broad range of disease areas, including oncology, autoimmune disorders, fibrosis, and monogenic blood disorders.The Opportunity
The Scientist I, In Vitro Pharmacology will play a key role in the pre-clinical pharmacology and toxicology team conducting in vitro studies to support in vivo tLNP-CAR product candidate testing, optimization and development for various indications including autoimmunity, fibrosis or oncology. . The candidate will be an instrumental team member focused on the generation of non-clinical data required for candidate selection. This Is an onsite role based in San Diego, CA.
Responsibilities & Duties:
  • Develop cell-based assays to characterize mammalian cells’ function across multiple programs.
  • Utilize multi-color flow cytometry to phenotype immune cells.
  • Engineer mammalian cells using techniques such as electroporation, viral delivery, and nanoparticle transfections.
  • Isolate PBMCs and T cells from Leukomas.
  • Maintain and implement good laboratory practice including an accurate online laboratory notebook system, instrument and reagent validation, and personal safety precautions.
  • Collaborate with the Research department to help guide product candidate optimization and nomination.
  • Work collaboratively in a cross-functional team environment
  • Organize and communicate data (written and verbal) and participate in project team meetings and cross-departmental team meetings.
Requirements/Qualifications:
  • Typically met with (a) a Ph.D. in a scientific discipline (or equivalent) and no previous industry experience; or (b) a bachelor’s degree (or equivalent) and 8 years industry experience.
  • Ph.D. degree preferred in Immunology, Cell Biology, or related field.
  • Previous experience in chimeric antigen receptor (CAR) production and evaluation is desirable.
  • Excellent aseptic/sterile techniques and prior experience culturing of mammalian cells, including cell lines, T cells, and PBMCs.
  • Experience with viral and non-viral gene transfer techniques to engineer mammalian cells.
  • Experience designing and executing assays to assess T cell function including cytotoxicity, activation, proliferation, and cytokine profiling.
  • Proficiency in flow cytometry and data processing
  • Experience with engineered T cells (e.g., CAR T cells) is desirable.
  • Experience designing and executing plate-based assays, including ELISA and Luminex/MSD
  • Experience with scientific data analysis software including GraphPad Prism, Tableau is desirable.
  • Ability to independently conduct studies while working efficiently as part of a team.
  • Ability to quickly learn new skills and acquire knowledge on the job.
  • Ability to manage multiple tasks, perform work with a high degree of attention to detail, and be part of an exciting fast-paced scientific environment.
  • Strong organizational and time-management skills, thorough record keeping skills (documentation and electronic lab notebook), excellent interpersonal, verbal, and written communication skills.
